## In brief

- Tango reports an 82% tumor response rate for its trial of a combination therapy in advanced pancreatic cancer, a result that matters to people living with the disease and their families [[1]](https://www.statnews.com/2026/10/23/tango-pancreatic-cancer-star-moderna-readout-newsletter/).
- The 82% response rate is a drop from the 92% response rate Tango reported in June, which came from fewer patients [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).
- Dose level 2, which used 250 mg vopimetostat and 600 mg daraxonrasib, was not well tolerated [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).

## The finding

Tango reports an 82% tumor response rate for its trial of a combination therapy in advanced pancreatic cancer, a result that matters to people living with the disease and their families [[1]](https://www.statnews.com/2026/10/23/tango-pancreatic-cancer-star-moderna-readout-newsletter/). The combination pairs vopimetostat with daraxonrasib [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).

CEO Malte Peters said the response rate and durability of the combination surpass any published PDAC datasets, even in front-line settings [[3]](https://www.tikr.com/blog/tango-therapeutics-sinks-13-on-pancreatic-cancer-data-it-says-surpass-any-published-pdac-datasets?ref=yahoofinance). PDAC is pancreatic ductal adenocarcinoma, the most common form of pancreatic cancer.

## Why the number changed

The 82% response rate is a drop from the 92% response rate Tango reported in June, which came from fewer patients [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3). In June, the company reported data from 12 patients showing a 92% response rate and 90% progression-free survival at six months [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).

Progression-free survival is the share of patients whose cancer has not grown at a set time point. The new figure comes from a larger group, so the two numbers are not drawn from the same pool of patients [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).

## Dosing results

Dose level 2, which used 250 mg vopimetostat and 600 mg daraxonrasib, was not well tolerated [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3). Three of four patients at that level experienced dose-limiting toxicities, meaning side effects serious enough to limit the dose [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).

## The drug behind the combination

The FDA approved daraxonrasib, as Rasonque, for second-line pancreatic cancer [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3). In September, weeks after that approval, the agency granted Breakthrough Therapy Designation to daraxonrasib in the first-line setting [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).

## What Tango plans next

Tango plans to launch a Phase 3 trial of the combination in front-line pancreatic cancer patients with MTAP deletions in the first half of 2027 [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3). Approximately 40% of patients with pancreatic cancer have an MTAP deletion [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).

The company also plans to seek FDA guidance on a possible accelerated approval path for vopimetostat plus daraxonrasib in second-line pancreatic cancer [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3). A Friday SEC filing says Tango is deprioritizing novel target discovery [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).

## What it means for you

If you or a family member has pancreatic cancer, the MTAP deletion is the detail to watch, because Tango's planned Phase 3 trial is aimed at front-line patients with that deletion [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3). About 40% of patients with pancreatic cancer have one [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).

Daraxonrasib is already FDA approved, as Rasonque, for second-line pancreatic cancer [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3). The combination with vopimetostat is not approved, and Tango plans to ask the FDA about a possible accelerated approval path in the second-line setting [[2]](https://www.biospace.com/drug-development/esmo-2026-tango-trips-on-sinking-response-rates-in-pancreatic-cancer-but-will-move-to-phase-3).
